Das Prostatakarzinom ist aktuell die zweith\u00e4ufigste Tumorerkrankung bei M\u00e4nnern weltweit und aufgrund der hohen Anzahl an Sterbef\u00e4llen ein Schwerpunkt der Tumorforschung. Patienten mit einem fortgeschrittenem Prostatakarzinom werden mit einer Androgen deprivierenden Therapie behandelt, die allerdings nach Langzeitbehandlung oft zu einem nicht mehr behandelbaren Tumorrezidiv f\u00fchrt. Ein Grund daf\u00fcr, ist die neuroendokrine Differenzierung von Prostatakarzinomzellen, welche Androgen-unabh\u00e4ngig die Tumorprogression f\u00f6rdert. Der Forschungsschwerpunkt von Dr. Wieseh\u00f6fer beruht auf der Untersuchung dieses Prozesses sowie der Eigenschaften der Neuroendokrin-\u00e4hnlichen Zellen. Hierbei m\u00f6chte er Gene und nicht-kodierende RNAs identifizieren, die einen potenziellen Einfluss auf die neuroendokrine Differenzierung haben, um m\u00f6gliche Therapieans\u00e4tze und neue Diagnostikverfahren zu entwickeln.Eine relativ neue Therapieform ist die Immuntherapie, welche der immunsupprimierenden Wirkung der Tumorzellen und der damit verbunden Immunevasion, entgegenwirkt. In Zukunft soll das Tumormikromilieu und die Mechanismen der Immunevasion von Prostatakarzinomzellen untersucht werden.<\/span>F\u00fcr dieses Ziel bedient sich Dr. Wieseh\u00f6fer einer Vielzahl von Methoden aus der Molekularbiologie, Proteinbiochemie und Zellbiologie. Zu den molekularbiologischen Methoden z\u00e4hlen Gen-Klonierungen und verschiedene Methoden der Expressionsanalyse. In der Proteinbiochemie werden immunologische F\u00e4rbungen von Geweben und Zellen angefertigt. Funktionelle Untersuchungen von Genen\/ Proteinen werden mit Hilfe verschiedener <\/span>in vitro<\/em> Analysen, wie z.B. der Durchflusszytometrie durchgef\u00fchrt.<\/span><\/p>\n

Prostate carcinoma is currently the second most common tumor disease in men worldwide and, due to the high number of deaths, a focus of tumor research. Patients with advanced prostate carcinoma are treated with androgen-depriviating therapy, which, however, often leads to an untreatable tumor recurrence after long-term treatment. One reason for this is the neuroendocrine differentiation of prostate cancer cells, which promotes androgen-independent tumor progression. The research focus of Dr. Wieseh\u00f6fer is based on the investigation of this process as well as the properties of neuroendocrine-like cells. Here, he wants to identify genes and non-coding RNAs that plays a potential role in neuroendocrine differentiation in order to develop possible therapeutic approaches and new diagnostic tools.Immunotherapy is a relative new treatment method, which counteracts the immunosuppressive effect of tumor cells and the associated immune evasion. In the future, the tumor microenvironment and the mechanisms of immune evasion of prostate carcinoma cells will be investigated.<\/span>For this aim, Dr. Wieseh\u00f6fer uses a variety of molecular biology, protein biochemistry and cell biology methods. Molecular biological methods include gene cloning and various methods of expression analysis. In protein biochemistry, immunological staining of tissues and cells is prepared. Functional investigations of genes\/proteins are performed with different <\/span>in vitro<\/em> analyses, e.g. flow cytometry.<\/span><\/p>\n
